Obviously a late reply, but I just figured it out as I had the same problem today. Just right click on the Behaviour program .exe and "run as administrator". That's it! If you're sure that your serial cable is working fine the it should work.
This is a really annoying part of Vista which has caused me problems many times. Generally, if a program in Vista doesn't seem to work right, try this option before choosing compatibility mode (which generally doesn't fix anything!).
